What does TaqI recognize?
TaqI recognizes the 4 bp sequence TCGA (5′→3′), cutting the top strand between position 1 and 2 within the site to leave a 5′ overhang of 2 bases.
What buffer should I use for TaqI?
rCutSmart gives the highest activity (incubate at 65°C). Values from TaqI-v2 (R0149), the only current NEB product: r2.1 and rCutSmart are both 100%, r1.1/r3.1 50%.
Does TaqI have star activity?
No documented star activity under standard reaction conditions.
Is TaqI blocked by DNA methylation?
Yes — blocked by Dam methylation. Use methylation-free template (e.g. a dam⁻/dcm⁻ strain) if this matters for your digest.
Can I heat-inactivate TaqI?
No — TaqI is not heat-inactivable. Use a column cleanup or phenol-chloroform extraction to stop the reaction instead.
